Ian McKellen pretty much was Gandalf.  His Magneto was cool and everything but nowhere near Gandalf.

James Gandolfini as Tony Soprano.  Pretty iconic.  When I see him in other roles I just couldn't buy it.

Jermey Piven as Ari Gold was another one who just became the character to me.

Eric Stoltz as Marty McFly.  Heh, they dodged a bullet there by recasting Michael J. Fox as the true perfect choice.

Lots of good choices ITT.

Bogart as Sam Spade in The Maltese Falcon and Sidney Greenstreet as Kaspar Gutman.
Whedon struck gold in Buffy the Vampire Slayer with Sarah Michelle Gellar and Alyson Hannigan. Plus the entire cast of Firefly and most of the supporting cast too.
Eastwood as Josey Wales.
Marisa Tomei in My Cousin Vinny.
