On May 10 in the journal Current Biology, researchers describe a recently identified bat coronavirus that is SARS-CoV-2's closest relative in some regions of the genome and which contains insertions of amino acids at the junction of the S1 and S2 subunits of the virus's spike protein in a manner similar to SAR-CoV-2. While it's not a direct evolutionary precursor of SARS-CoV-2, this new virus, RmYN02, suggests that these types of seemingly unusual insertion events can occur naturally in coronavirus evolution, the researchers say.

"Since the discovery of SARS-CoV-2 there have been a number of unfounded suggestions that the virus has a laboratory origin," says senior author Weifeng Shi, director and professor at the Institute of Pathogen Biology at Shandong First Medical University in China. "In particular, it has been proposed the S1/S2 insertion is highly unusual and perhaps indicative of laboratory manipulation. Our paper shows very clearly that these events occur naturally in wildlife. This provides strong evidence against SARS-CoV-2 being a laboratory escape."

The researchers identified RmYN02 from an analysis of 227 bat samples collected in Yunnan province, China, between May and October of 2019. "Since the discovery that bats were the reservoir of SARS coronavirus in 2005, there has been great interest in bats as reservoir species for infectious diseases, particularly as they carry a very high diversity of RNA viruses, including coronaviruses," Shi says. RNA from the samples was sent for metagenomic next-generation sequencing in early January 2020, soon after the discovery of SARS-CoV-2.

Across the whole genome, the closest relative to SARS-CoV-2 is another virus, called RaTG13, which was previously identified from bats in Yunnan province. But RmYN02, the virus newly discovered here, is even more closely related to SARS-CoV-2 in some parts of the genome, including in the longest encoding section of the genome called 1ab, where they share 97.2% of their RNA. The researchers note that RmYN02 does not closely resemble SAR-CoV-2 in the region of the genome that encodes the key receptor binding domain that binds to the human ACE2 receptor that SARS-CoV-2 uses to infect host cells. This means it's not likely to infect human cells.

The key similarity between SARS-CoV-2 and RmYN02, is the finding that RmYN02 also contains amino acid insertions at the point where the two subunits of its spike protein meet. SARS-CoV-2 is characterized by a four-amino-acid insertion at the junction of S1 and S2; this insertion is unique to the virus and has been present in all SARS-CoV-2 sequenced so far. The insertions in RmYN02 are not the same as those in SARS-CoV-2, which indicates that they occurred through independent insertion events. But a similar insertion event happening in a virus identified in bats strongly suggests that these kinds of insertions are of natural origin. "Our findings suggest that these insertion events that initially appeared to be very unusual can, in fact, occur naturally in animal betacoronaviruses," Shi says.

The cave, whose exact location is being kept secret, is inhabited by wild bats that have been found to carry a  “rich gene pool of SARS-related coronaviruses,” said Peter Daszak, the president of EcoHealth Alliance, a U.S. nonprofit organization that monitors wildlife diseases that could pose a pandemic risk.

Daszak said that one of the 500 or so virus strains discovered in 2004 is 96 per cent similar to the novel coronavirus that has infected 28,000 people and killed 560 since the outbreak began in December.  

“What we’re saying is that this cluster of viruses is a high risk,” Daszak said.

The cave was discovered as part of the team’s efforts to track viruses similar to SARS in 2003, after the epidemic had struck, Daszak explained. At the time, people believed civets had caused the outbreak, but Daszak’s team disputed that notion.

Bats host a much higher number of zoonotic viruses than other mammals, many of which have caused human disease and outbreaks. A 2019 study warned that bats could cause the next coronavirus epidemic in China, due to their geographic proximity to several urban hotspots. This cave, for example, is located 60 kilometres from the city of Kunming in the Yunnan province of China.

After the SARS epidemic, the team did not find many bats in the wildlife market, Daszak said, but noticed that people were “hunting them in the wild and selling directly to restaurants.” It collected faeces from bats that lived in the cave and tested them.

“We found viruses in bats that could infect human cells in a lab,” he said.  

The team then tested the viral strains on mice to see whether they would cause a SARS-like disease. They did.

Finally, the team drew samples from people who lived near the bat caves and found that three per cent of them had developed antibodies to the viruses — proving that the strains can and have infected humans in the past.

“So that was a red flag,” Daszak said.

But a lack of funding prevented scientists from researching the virus now known to be a close relative of the 2019 novel coronavirus.

“It’s not that we didn’t find it interesting,” Daszak said. “It went back into the freezer because we didn’t have enough money.”

Instead, the researchers focused on the strains that most resembled SARS and flagged those as most high risk to authorities.

The team published its results in a number of accredited academic journals and Daszak argued for the viruses to be added to the WHO’s top pathogens of high risk to human health.

After flagging the viruses, the team went back to the same regions in China to interview people in rural communities and learn who was at highest risk in contact with wildlife. “We published that information, too, and now if you go to some of those caves, there’s a sign up that says don’t go in, no entry. That’s the sort of thing that the government does,” he explained.

This cave, he said, has since been redeveloped but declined to elaborate.

“(WHO) took it seriously. The Chinese government took it seriously,” Daszak said. China has attempted to ban the trade and has even banned consumption of wildlife from government banquets, which is “a big thing,” he added.

However, it’s tough to block all the ways, such as the wildlife trade, that these viruses “spill over” into human populations. Behaviours like these, he explained are “culturally deep in our population habits.… These things go back 5,000 years of history, it’s not straightforward. You can’t just ban it and it goes away.”

Scientists studying the current coronavirus outbreak have not yet narrowed down how the disease spread to humans. If there was enough funding, however, Daszak believes that further research on the strain might have made a difference to preventing or minimizing the impact of the current outbreak. The funding could have supported efforts to sequence the virus genome, test it on animal models and then, depending on results, flag it as a “high risk factor” to respective authorities.

“You can’t say for sure” whether the current pandemic would have been prevented, he stressed. “We also don’t know for sure that blocking, you know, talking to communities and trying to get this reduced contact would actually stop an outbreak. But every little bit helps,” he said.

There also isn’t enough large-scale support or funding to finance efforts to prevent pandemics from occurring in the future — a long-term, yet realistic endeavour, said Daszak.

Last year, PREDICT, a U.S. federal program to identify wildlife viruses that could infect humans was shut down by the government due to “the ascension of risk-averse bureaucrats,” according to Dennis Carroll, the former director of the United States Agency of International Development (USAID).

Over the past decade, the initiative had discovered more than 1,000 new viruses, including a new strain of Ebola. It also trained people and created medical infrastructure in several developing countries to prepare for potential outbreaks.

USAID also funded the $200 million Global Virome Project (GVP), an international effort launched in 2016 to identify and catalogue 99 per cent of “all zoonotic viruses with epidemic/pandemic potential.”

GVP estimates that there are about 1.5 million viruses present in wildlife, some of which could pose a risk to human health. The organization needs between $1.2 billion and $3.4 billion to find them — a minimum of $125 million a year.

It looks like a big number. Yet, CNN reported that the coronavirus outbreak could cost China $60 billion in lost economic growth. After SARS broke in 2003, an estimated $40 billion was lost in productivity.

“It’s hard to maybe fight for something that you don’t know about when there are other things that are killing people,” said Goldstein, a virologist at the University of Carolina, Davis who works with the GVP. “But examples like these just explain why we need to continue investing in both of these streams.”

While “it’s hard to say what will prevent an outbreak,” once authorities can understand where and when people are coming into contact with a potentially risky virus, and the kind of population behaviours that initiate contact, “you can think about how to prevent that or how to reduce contact,” she said.

Federal Judge Blocks North Carolina Governor's Restrictions on Church Services

On Saturday, a federal judge temporarily blocked North Carolina Democratic Gov. Roy Cooper's restrictions on indoor religious services during the Wuhan coronavirus pandemic. In a recent statewide stay-at-home order aimed at reopening the state, religious services were instructed to be held "outdoors unless impossible."

Judge James C. Dever III sided with two Baptist churches who argued Gov. Cooper's restrictions on indoor church services violate their rights to worship and treat churches differently than secular entities like retailers.

While Dever believes the governor has acted in good faith, the judge also finds restrictions applied to one group and not another burden religious freedom and do little to stop the spread of the coronavirus, the judge wrote in a 22-page order.

"The record, at this admittedly early stage of the case, reveals that the Governor appears to trust citizens to perform non-religious activities indoors (such as shopping or working or selling merchandise) but does not trust them to do the same when they worship indoors together," Judge Dever wrote in his order.

A spokesman for the governor, Ford Peter, said the governor will not appeal the decision despite disagreeing with the judge's ruling, according to The Washington Times. Judge Dever set the next hearing for May 29.

I think Q first said to watch the water Feb. 15, 2018.  Interesting to note a CDC researcher disappeared Feb. 12, 2018 and was later found dead in the water.

Rick Grennell, Acting DNI (and ambassador to Germany) who has been responsible for getting all this stuff declassified and much more. He is a Team Trump super hero these days.

ETA: He is a cancer survivor, not sure what type so I don't know if it involved his eye.

Chinese ambassador to Israel found dead in Herzliya home

There are reportedly no signs of violence. Wei's death comes a week after US Secretary of State Mike Pompeo visited Israel in an apparent attempt to convince the gov't to limit Chinese investment.

The Chinese Ambassador to Israel, Du Wei, was found dead in his Herzliya home on Sunday morning, a Foreign Affairs Ministry official confirmed. Police are currently in his home and investigating.

The Chinese embassy, however, said that it cannot confirm the reports as of yet.

Arabs boycott Facebook after Muslim Brotherhood sympathizer Tawakkol Karman joins content board

Yemeni journalist and political activist Tawakkol Karman has complained of “widespread bullying and smear" after Facebook’s decision to induct her into its content Oversight Board plunged her into controversy. But what if her credibility was more at risk from her own words and actions than any  alleged “smear campaign?”

To much of the globe, Tawakkol Abdel-Salam Khalid Karman is the first Arab woman — and the second Muslim woman — to win a Nobel Prize, in 2011.

Less well known is the fact that Karman held a senior position with her country’s Al-Islah Party, an affiliate of the Muslim Brotherhood known regionally for its divisive and violent agenda.

Karman has severed ties to the Brotherhood’s Yemeni branch, an Islamist movement founded by Abdul Majeed Al-Zindani, a man who figures in Washington’s Specially Designated Global Terrorist list. But many wonder whether the move was merely a cosmetic exercise.

Unsurprisingly, Facebook’s choice has prompted outrage on social media networks, with many worried that it will bring the Muslim Brotherhood’s ideas right into the heart of the biggest social networking company in the world.

“She has not denounced the extremist ideology of the Muslim Brotherhood,” Ghanem Nuseibeh, founder of risk consultancy Cornerstone Global Associates, told Arab News.

“On the contrary, there is everything to believe that she continues to espouse the hate speech that has been a mark of the Brotherhood in general.”

Given her prominent role in the revolution that toppled Yemen’s former leader Ali Abdullah Saleh, Karman’s Nobel Prize is not without merit, say political analysts. But they add that her advocacy of extremist causes can hardly be glossed over.

“Karman was considered a symbol of the Yemeni revolution against the rule of Saleh, but over time she has become associated with intolerance, discrimination and lack of neutrality,” Hani Nasira, a terrorism and extremism expert, told Arab News.

Soon after Karman was awarded the Nobel Prize, she was invited to Doha and personally congratulated by Yusuf Al-Qaradawi, the Muslim Brotherhood leader and preacher of hate, whose fatwas call for suicide bomb attacks and who praises Hitler for “punishing” the Jews.

After conveying to her his message of “support” for the Yemeni people, Al-Qaradawi gave Karman a copy of his book, “Fiqh Al-Jihad,” as a gift.

Atwater city officials defy Cali Gov. Newsom's orders.

California City Declared a ‘Sanctuary’ From Stay-Home Order

More parks and hiking trails welcomed visitors again Saturday and one city declared itself a “sanctuary” from California's stay-at-home order as the state's diverse regions carve their own path toward reopening.

Officials in Atwater, a city of 30,000 in central California, unanimously agreed Friday not to enforce the nearly 2-month-old order intended to slow the spread of the coronavirus, meaning local authorities won't interfere with any business or church that decides to reopen ahead of state restrictions.

The declaration was a symbolic gesture of defiance against Gov. Gavin Newson's order and the city's mayor cautioned that businesses were taking their own risks by reopening.

"If you do have a state (business) license, that’s between you and the state of California,” Mayor Paul Creighton told the Merced Sun-Star.
