In Harm's Way - Duke's acting even impressed Premeinger.

The Commancheros - Duke and Lee Marvin. Awesome.

The Alamo - Historically laughable but the Flaca speech should be required ARF viewing.

True Grit - Duke's only Oscar. So many quoteable lines. "I always go backwards when I'm backing away!!"

The Quite Man - My Irish grandmother made me watch this every St. Patrick's Day.
Duke, John Ford and a great cast firing on all cylinders.

Hard to have a favorite, when he only has the one character.

But, I always liked The Horse Soldiers & Rio Bravo.

And True Grit goes without saying I suppose.

Only one character?  I don't know about that.  But......just watch John Wayne movies, doesn't matter which one.  Pay attention.  Think about what you saw on screen after the movie's over.  Compare/contrast with the world you see you around you.

And realize that there was a time when most people grew up watching John Wayne movies and thought that what you saw on the screen is completely normal and reasonable.
