You can do either way but if you do it with a router (assuming you have one) a simple vice is all that is needed with the jig.
A drill press would require a good x-y vise. Plus with a router it is easier to access the cavity to vacuum out the chips and to look down and see your progress |

A router will give you a far superior finished product.
Drill presses are not designed to cut latterly only vertically. Lack of good horizontal support on the spindle/quill/chuck causes substantial chattering. Routers and mills are specifically designed to handle lateral loading and will give you a much nicer finish. |

I used a regular drill for the holes before routing as the jig is pretty high and would not fit under my cheap drill press
I did protect the sides with blue painters tape just in case but I did not see any marks on it when removing. Better safe than sorry was my motto |
