I am installing an IWC QD End Plate on my AR-15.  Is it necessary to stake the castle nut as suggested in a few instructional videos?  Would "thread locker" be an effective alternative?

Thread locker can make it a bear to remove in the future.  I would skip using any.  
If you torque it to the recommended spec of around 35 ft/lbs, you'll be fine.  
Staking is optional, but really you're just moving a small piece of metal from the end plate into the notch to keep it from ever backing off.  
2 minutes with a sharp punch, nail, or drywall screw and a few taps with a hammer and you're all done. Cheap insurance.

Thanks.  I did not know the torque spec and was glad you posted it.  Since the end plate was not staked at the factory, I will skip that step for now.    I agree that thread locker may cause the whole assembly to rotate and it's best not to use any.   I only use the firearm at the range so it will be minimal  risk if the castle nut loosens.

Any loosening of the castle nut is easily noticed.
Routine maintenance and cleaning should include a quick visual of the entire extension.

Or just use a very small amount of Blue loctite.

Ya, no harm in this I wouldn't think. After all, I had to get my castle nut off on an old rifle and RRA used red loctite on it.... that wasn't fun.

Just in case something goes wrong, I'd just torque it for now. Then if you realize the new endplate has an issue or something, easier to deal with until you know it's square. You can always recheck torque quick and stake later, once it's all confirmed good.
