Hello,
I efilled a Form 1 on 1/10 and its current state is "Submitted / In Progress". I was originally going to build a 9mm SBR with a 4.5 barrel. Now, I have changed my mind and want to build a 7.5 5.556 rifle. Should I just wait till it is approved then send the ATF a written letter? Do I need to wait for a response?

Do nothing until you get the Form, try to keep things simple for ATF.

Then, if you want to, send them the letter.  There's no legal requirment to do so.
