My glocks guide rod keeps on slipping off the first notch ( the one closest to the barrel) and resting on the one farthest from the barrel. Is it supposed to do this? Any bad affects from this?

Cycle the slide back a few 3-5 times. If there is any binding have a Glock armorer look at it. Mine (23)doesn't ride on the closest one either.

Check the end of the rod tab (the flat round part that rests in the notch on the barrel) and make sure it isn't cracked or deformed in any way.  If anything is wrong with it, it may not stay put where it should be.  If it is, Glock will replace it.  It happened with mine once, but never again in 15 years.

The "circular cut" closest to the barrel is for assemble purposes only. When the slide is installed the guide rod & spring rest against the slide lock lever. It is normal for the guide rod to ride up out of the notch during slide removal. Make sure when you install the slide that the guide rod assemble is seated fully in the circular cut, otherwise you might chip, crack or break the end of the guide rod assembly.

As he said, it should do that and be re-seated everytime. It's not only normal, it's needed for the gun to work. Oh note: I'm a certified Glock Armorer, not just running at the mouth.
