I bought some nylon inserted locking nuts to replace the knobs on my carry handle. The knobs reach a certain point and then they won't come off. Do I just get after it and keep twisting them off with brute force, or do I need to drill it out? Or is there some other way?

The last thread is damaged on purpose on some (yours).
Brute force might twist the screw in two.
Reaching in there with a (gasp) Dremel might let you take it off.
Your call.

That's what I figured. I think I'll get a drill after it. I just didn't want to miss something hidden that would keep me from tearing anything up.

It depends on how it was inserted. It might have a nut that is attached to it that's counter-acting at the end or it may be smashed in place.

While not a carry handle, I did once had the acog mount get stuck into place and couldn't remove it. What I did was used a hacksaw to cut the knob down. It took me about an hour or so and a lot of aluminum debris came off but I finally got it off. Luckily I was replacing the mount so it didn't matter that it broke.

The threaded rod ends are staked.

There was a thread on replacement of these with another nut. Looked real good.

Yep, the ends look like a shallow Phillips head screw.
